It's been pointed out before that this rule isn't always enforced. I have missed a question before myself and nothing happened I am pleased to say. It could be helpful to add a notice to your listings and/or your about me page that you are not around at weekends.

I answer questions every day, so I never thought about this rule. I can see how much of an issue it can be for a lot of sellers though. My answer isn't open up on Saturdays and Sundays. You can sign into your eBid from any computer, I don't expect you'll get suspended here for using a different computer than your normal one like ebay; and then you can just answer emails once each day to get you through. That is my solution until their is a rule change; which I think is ultimately the best move. Something similar to the rule as it stands, maybe have a settings to either accept emails on weekends, or not to. If you don't anyone sending you mail during the weekend could get a message from eBid stating that you will not receive the mail until Monday, and not to expect a reply until then.
This could also be a problem when the seller takes a vacation. Even if the holiday notice is up and no one can buy, they can still post questions. I expect support looks carefully before punishing someone for breaking the 24 Hour Rule. I think this would be done manually on a case-by-case basis.
